    一用途

    叶绿素含量测量仪广泛应用于植物生理生态研究,特别是对传统吸收法测量困难的小叶片或样品的叶绿素含量进行测量,例如针叶、发育未*的水稻、生于岩石上的丝状藻、地衣、草坪草、仙人掌、龙舌兰属植物、菠萝、拟南芥、果实、苔藓、叶茎、叶柄。

    二原理

    尽管实践已经证明了吸收技术测量叶绿素含量的有效性,但该方法对样品的要求较高,使其应用范围受到限制。而样品本身的叶绿素荧光则不受样品厚度、均一性等的影响,可以指示叶绿素的含量。根据这个原理,Gitelson等比较了叶片的吸收和反射对叶绿素荧光的影响,发现在735nm和700nm处的比值与叶片的反射相关性最高,并且当样品叶绿素含量在41mg·m-2到675mg·m-2时,F735/F700的比值与其叶绿素含量间存在着显著的线性关系(R2≥0.95),与传统的吸收性测量方法相比(测量范围≤300mg·m-2),提高了叶绿素含量的线性关系范围。

    根据Gitelson的研究结果,CCM-300使用具有15nm半峰宽的460nm二极管用于荧光激发,提高了红色荧光波长,避开了大约685nm的发射荧光峰(叶绿素可重新吸收,降低测量精度)。仪器使用在735nm和700nm的叶绿素发射荧光的比率(F735/F700)进行叶绿素含量的测量,读数为F735/F700的比值以及根据Gitelson方程计算的叶绿素含量。
